目的:掌握河源地区血小板捐献者HLA Ⅰ类(-A、B位点)和HPA基因信息資料,初步建立本地区血小板基因信息库。方法:选择2017年9月6日至2020年9月6日期间在本地区捐献血小板的200例固定献血者为研究对象,采用PCR-SSP技术对其血液样本进行HLA Ⅰ类(-A、B)基因和HPA1-17系统基因分型,分析其基因频率分布情况。结果:200例样本中共检出HLA-A、B位点等位基因41种。其中HLA-A位点检出15个,HLA-B位点检出26个。HLA-A、B位点基因频率>0.05的是:A*11(0.2625)、A*2(0.2225)、A*33(0.1775)、A*24(0.1175)、A*1(0.0650)和B*58(0.1425)、B*62(0.1400)、B*46(0.1225)、B*13(0.0750)。HPA1-17系统基因分型中等位基因HPA15a、HPA3a、HPA5a、HPA2a、HPA1a呈多态性分布,其频率分别为:0.555、0.570、0.950、0.968、0.990。呈单特异性分布的等位基因是:HPA4a、HPA6a-14a,HPA16a、HPA17a,a基因频率均为1,b基因频率为0。对偶抗原不配合率前二位分别为HPA-15(37.20%)和HPA-3(37.00%)。结论:初步建立河源地区血小板捐献者HLA-A、B位点与HPA1-17系统基因信息库。 |

Abstract: |
Objective:To establish genetic database of HLA class I(-A,B locus)and HPA about platelet donors in Heyuan area. Methods:200 regular blood donors who donated platelets in the region from September 6,2017 to September 6,2020 were selected as research subjects. Polymerase chain reaction sequence specific primer(PCR-SSP)applied in genotyping for HLAclass I(-A,B locus),and HPA1-17,Analyze its gene frequency distribution. Results:Total of 41 HLA-A and B alleles were detected in 200 apheresis platelets donors. There were 15 alleles at HLA-A locus and 26 alleles at HLA-B locus. The gene frequencies of HLA-A、B> 0.05 were:A * 11(0.2625),A * 2(0.2225),A * 33(0.1775),A * 24(0.1175),A * 1(0.0650),and and HLA-B wereB * 58(0.1425),B * 62(0.1400),B * 46(0.1225),B* 13(0.0750). The alleles with polymorphic distribution in the genotyping of HPA1-17 system were HPA15a,HPA3a,HPA5a,HPA2a and HPA1a,and their frequencies were 0.555,0.570,0.950,0.968 and 0.990 respectively. The alleles with monospecific distribution of that were HPA4a,HPA6a-14a,HPA16a and HPA17a,with Agene frequency of 1 and B gene frequency of 0. The first two incompatibility rates were HPA-15(37.20%)and HPA-3(37.00%). Conclusion:The gene information database of HLA-A,B locus and HPA1-17 system was preliminarily established in Heyuan area . |
